Resumo Os sorvetes à base de frutas são produtos amplamente consumidos em países tropicais e, devido à sua composição, podem ser uma boa fonte de crescimento microbiano, incluindo patógenos oportunistas. Os objetivos deste estudo foram caracterizar as populações de leveduras presentes nos sorvetes brasileiros à base de frutas e investigar sua susceptibilidade aos antifúngicos anfotericina B, fluconazol e itraconazol, e a capacidade dos isolados, que puderam crescer a 37 °C, de aderir em células epiteliais bucais. Duzentos e sessenta e sete isolados de levedura obtidos das amostras de sorvete foram identificados como pertencentes a 29 espécies, com contagens variando de 1,5 a 5,2 log UFC/mL. As espécies predominantes foram Candida intermedia, Torulaspora delbrueckii, C. parapsilosis, Clavispora lusitaniae, Saccharomyces cerevisiae e Pichia kudriavzevii. Pelo menos 16 espécies de leveduras isoladas neste estudo foram relatadas como agentes patogênicos oportunistas. Quarenta e um isolados de leveduras apresentaram resistência ou susceptibilidade dose-dependente a pelo menos um dos antifúngicos testados. Um isolado de C. parapsilosis foi resistente a todos os antifúngicos testados e mostrou capacidade de aderir a células epiteliais bucais. A porcentagem de adesão foi alta principalmente para isolados de P. kudriavzevii, Meyerozyma guilliermondii, C. parapsilosis, S. cerevisiae e Debaromyces hansenii. Os dados sugerem que a presença dessas leveduras oportunistas como contaminantes nos sorvetes pode representar um risco para o consumidor final, especialmente para indivíduos imunocomprometidos que podem consumir esses produtos.

Abstract Fruit-based ice creams are products widely consumed in tropical countries and, because of their composition, can be a good source for microbial growth, including opportunistic pathogens. The aims of this study were to characterize the yeast populations present in Brazilian fruit-based ice creams, and to investigate the antifungal susceptibility to amphotericin B, fluconazole and itraconazole, and the ability of the isolates that were able to grow at 37 ºC to adhere to buccal epithelial cells (BEC). Two hundred and sixty-seven yeast isolates obtained from the ice cream samples were identified as belonging to 29 species, with counts that ranged from 1.5 to 5.2 log CFU/mL. The predominant species were Candida intermedia, Torulaspora delbrueckii, C. parapsilosis, Clavispora lusitaniae, Saccharomyces cerevisiae and Pichia kudriavzevii. At least 16 yeast species isolated in this study have been reported as opportunistic pathogens. Forty-one yeast isolates showed resistance or dose-dependent susceptibility to at least one of the antifungal drugs tested. One isolate of C. parapsilosis was resistant to all the antifungals tested and showed ability to adhere to BEC. The percentage of adhesion to BEC was high mainly for isolates of P. kudriavzevii, Meyerozyma guilliermondii, C. parapsilosis, S. cerevisiae and Debaromyces hansenii. The data suggest that the presence of these opportunistic yeasts as contaminants in ice creams may represent a potential risk to the final consumer, especially to immunocompromised individuals who may consume these products.

RESUMO: O interesse no chá verde comercial (Camellia sinensis) (L.) Kuntze (Theaceae) tem aumentado nos últimos anos devido suas propriedades medicinais. No presente estudo utilizamos a microextração em fase sólida, em combinação com a cromatografia gasosa acoplada com espectrometria de massas, como um eficiente método de caracterização de compostos voláteis e efetuamos o controle de qualidade de C. sinensis. Comparamos também a anatomia das folhas de diferentes amostras comerciais de chá verde, as informações de seus rótulos e determinamos a presença de matérias estranhas, de microrganismos, o teor de umidade e de cinzas totais. Entre as trinta amostras analisadas, seis foram identificadas como C. sinensis de acordo com o estudo anatômico; 83,4% não estavam de acordo com os limites especificados para matérias estranhas e 6,6% para o teor de umidade. Todas as amostras apresentaram cinzas totais de acordo com a legislação vigente. As análises evidenciaram diversos compostos orgânicos voláteis distintos, sendo majoritários terpenos, hidrocarbonetos e álcoois. As análises microbiológicas mostraram que 13,3 e 16,6% das amostras excedeu os limites de contagem para bolores e leveduras, e para bactérias mesófilas, respectivamente. Por fim, 73,3% das amostras apresentou Escherichia coli acima dos limites sanitários estabelecidos.

ABSTRACT: The interest in the use of green tea (Camellia sinensis) (L.) Kuntze (Theaceae) products have increased in the last few years due to its medicinal properties. In the present study, we proposed that headspace solid phase microextraction, in combination with gas chromatography coupled to mass spectrometric could be an efficient method to assess the volatile compounds and to ensure the quality control of C. sinensis. We have also compared the anatomical leaf of different commercial green tea samples, analyzed the information in their labels and determined the presence of foreign materials, moisture content, total ashes and microorganisms. Among the 30 analyzed samples, six were identified as C. sinensis according to the anatomical study; 83.4% were not in accordance with the specified limits for foreign matter and 6.6% for moisture content. All samples presented total ashes in accordance with the current legislation. The analysis indicated the presence of several different volatile organic compounds, being terpenes, hydrocarbons and alcohols the major ones. Microbiological analysis showed that 13.3% and 16.6% of the samples exceeded the limits for counts of molds and yeasts, and for mesophilic bacteria, respectively. 73.3% presented E. coli above the established sanitary limits.

ABSTRACT Chicha, a type of beer made mainly with maize or cassava, is a traditional fermented beverage of the Andean region. There have only been a few studies on yeasts associated with chicha fermentation, and the species diversity occurring during the production of this beverage is not known. The objective of this study was to determine the biodiversity of yeasts in chicha, and to characterize the Saccharomyces cerevisiae populations associated with the production of chicha de jora, seven-grain chicha, chicha de yuca, and chicha de morocho in Ecuador. The molecular diversity of S. cerevisiae populations was determined by restriction polymorphism mitochondrial profiles. The beverages were characterized based on their physicochemical parameters. Twenty-six species were identified, and the most prevalent species were S. cerevisiae and Torulaspora delbrueckii. Other yeast species were isolated at low frequencies. Among 121 isolates of S. cerevisiae, 68 different mtDNA molecular profiles were identified. These results showed that chichas are fermented by a high number of different strains of S. cerevisiae. Some other species provided a minor contribution to the fermentation process. The chicha presented generally similar physicochemical parameters to those observed for other traditional fermented beverages, and can be considered as an acid fermented beverage.

Resumo A indústria vinícola brasileira apresentou crescimento significativo nos últimos anos e a inserção de conceitos ‒ como, por exemplo, indicações geográficas como sinais de qualidade ‒ colocou o Brasil em sintonia com as tendências da produção mundial. Este trabalho objetivou aplicar a técnica de microextração em fase sólida em combinação com a cromatografia gasosa-espectrometria de massa, para analisar vinhos brasileiros produzidos com diferentes variedades de uva, a fim de separar e identificar seus compostos orgânicos voláteis. Estes foram identificados por meio de comparações entre os espectros obtidos e os da base de dados da biblioteca do National Institute of Standards and Technology (NIST), e por comparações com índices de retenção linear e dados da literatura. A quantidade de compostos baseou-se nas áreas de pico total de cromatogramas. Quarenta e sete compostos voláteis foram identificados e agrupados em álcoois, aldeídos, ácidos graxos, ésteres, hidrocarbonetos, cetonas e terpenos. A maioria pertencia à função éster, conferindo aroma frutado aos vinhos. Os álcoois podem ter se originado a partir do metabolismo da levedura, contribuindo para aromas alcoólicos e florais. Lactato de etila, 1-hexanol e maleato de dietila foram identificados em todas as variedades de vinho, exceto Merlot. Decanal, citronelato de metila, 3-metilbutirato de (E)-2-hexenila foram encontrados apenas em Merlot, ao passo que 2,3-butanodiol, somente em vinhos Tannat. O composto 2-Feniletanol foi encontrado em todas as amostras e é reconhecido por conferir agradáveis aromas de rosa e mel aos vinhos. Este estudo evidenciou que o perfil volátil dos vinhos tintos é caracterizado, principalmente, por ésteres e álcoois superiores. Análises estatísticas de comparação de médias mostraram uma maior quantidade de médias significativamente distintas das áreas relativas para o vinho Merlot. A análise de componentes principais mostrou que um grupamento foi formado apenas por amostras de vinho Merlot, o que está provavelmente relacionado à existência de compostos orgânicos voláteis que foram, especificamente, identificados nestes vinhos.

Abstract The Brazilian wine industry has shown significant growth in recent years and the insertion of new concepts, such as geographical indications as signs of quality, has placed Brazil in tune with the tendencies of world wine production. The aim of this work was to apply the Solid Phase Microextraction technique in combination with Gas Chromatography-Mass Spectrometry to study Brazilian wines made from different grape varieties, in order to separate and identify their volatile organic compounds. These substances were identified by comparisons between the spectra obtained with those presented in the NIST library database, and by comparisons with linear retention indices and literature data. The amounts of the compounds were calculated based on the total peak areas of the chromatograms. Forty-seven volatile compounds were identified and grouped into alcohols, aldehydes, fatty acids, esters, hydrocarbons, ketones and terpenes. Most of them belonged to the ester function, conferring a fruity aroma on the wines. The alcohols may have originated from the yeast metabolism, contributing to the alcoholic and floral aromas. Ethyl lactate, 1-hexanol and diethyl maleate were identified in all the varieties, except Merlot. Decanal, methyl citronellate, (E)-2-hexenyl-3-methylbutyrate were only found in Merlot, while 2,3-butanediol was only present in the Tannat wines. 2-Phenylethanol was present in all varieties and is recognized as giving pleasant rose and honey attributes to wines. This study showed that the volatile profile of red wines is mainly characterized by esters and higher alcohols. The statistical analysis of the comparison of averages showed a greater amount of averages significantly different in the relative areas of Merlot wine. The Principal Component Analysis showed one grouping composed only of the Merlot wine samples, and this was probably related to the existence of the volatile organic compounds that were specifically identified in these wines.

A exposição humana à água contaminada constitui um importante mecanismo para a transmissão de patógenos gastrointestinais. Cepas de Escherichia coli diarreiogênicas podem causar infecções entéricas em humanos e incluem seis categorias diferentes de acordo com fatores de virulência. Este trabalho tem por objetivos detectar a presença de cepas diarreiogênicas de E. coli nas águas de sete praias do reservatório de Lajeado, Tocantins, Brasil e testar a resistência a drogas antimicrobianas para correlacionar com a possível contaminação da água por fezes humanas. As contagens de coliformes totais e E. coli foram realizadas pela técnica do substrato cromogênico ColilertTM. A identificação bioquímica foi realizada utilizando kit API20E e a detecção dos fatores de virulência por PCR, empregando primers específicos para Shiga, LT e ST e genes de intimina. A susceptibilidade a 16 antibióticos foi testada pela técnica de difusão em discos de papel impregnados com antibiótico. Entre as 149 cepas de E. coli testadas, duas cepas de EPEC e duas de ETEC foram detectadas em águas de praias situadas em áreas urbanas, perto de descarga de efluentes. Estas cepas apresentaram resistência de 3 a 6 antibióticos. A provável origem humana das cepas é sugerida devido ao perfil de multiresistentes apresentado.

The exposure to contaminated water constitutes an important mechanism for the transmission of gastrointestinal pathogens. Diarrheagenic Escherichia coli strains cause enteric infections in humans and include six different categories according to virulence factors. This paper aims at detecting the presence of diarrheagenic E. coli strains in bathing waters of seven beaches of Lajeado Reservoir, in the Tocantins River in Brazil, and to test the resistance to antimicrobial drugs to correlate with possible contamination of the water with human feces. Total coliform and E. coli counts were done by the ColilertTM chromogenic substrate technique. Biochemical identification was accomplished by API20E and detection of virulence factors by PCR, employing specific primers for Shiga, LT, and ST and intimin genes. The susceptibility to 16 antibiotics was tested by disk-diffusion technique. Among one hundred and forty-nine strains of E. coli, two strains of EPEC and two of ETEC were detected in waters of beaches situated in urban areas, close to sewage discharge. These strains presented resistance to three to six antibiotics. Human origin is suggested based on the multiresistant profile of these strains.

Duas linhagens de Saccharomyces cerevisiae foram testadas como iniciadoras em uma destilaria de cachaça. Foram utilizadas as linhagens de S. cerevisiae UFMG-A829, isolada de fermentação de cachaça, e S. cerevisiae K1-V1116, de origem vinícola. A permanência de cada linhagem durante a fermentação foi determinada por RAPD (Random Amplified Polymorphic DNA)-PCR, utilizando o iniciador M13. As duas linhagens predominaram nas dornas de fermentação por aproximadamente 30 dias. Leveduras não-Saccharomyces e S. cerevisiae indígenas foram isoladas em menor proporção durante o experimento. As linhagens de S. cerevisiae indígenas apresentaram perfis moleculares distintos em relação às linhagens iniciadoras. As duas linhagens foram promissoras para serem utilizadas como iniciadoras do processo fermentativo para a produção da cachaça.

Two Saccharomyces cerevisiae strains were tested as the starter yeasts in a traditional cachaça distillery. The strains used were S. cerevisiae UFMG-A829, isolated from a cachaça fermentation process, and S. cerevisiae K1-V1116, obtained from the wine industry. The permanence of each strain in the fermentation must was determined by RAPD (Random Amplified Polymorphic DNA)-PCR, with primer M13. Both yeast strains were prevalent in the vats for approximately 30 days. Indigenous non-Saccharomyces and indigenous S. cerevisiae strains were isolated in lower counts during the fermentation period. Indigenous S. cerevisiae strains were molecularly distinct when compared to the starter yeasts. The two yeasts appeared promising starter yeasts in the fermentation process to produce traditional cachaça.
